By blocking a the detection the undercritical emission (UAF), only the SAF emission is collected to reconstruct the confocal (or the STED) image. Only the detection of the microscope is modified to introduce the filtering of the UAF, and as can be seen on the images of microtubules, deeper ones disappear in the SAF image compared to the confocal image.
